Description

Three distinct isoforms of nitric oxide synthase (NOS), nNOS/NOS I, iNOS/NOS II, and eNOS/NOS III, catalyze the oxidation of arginine to citrulline and NO with Nω-hydroxy-L-arginine (NOHA) formed as an intermediate.1,2,3 N-HFG is an NOHA analog that serves as an alternate substrate for iNOS. It is oxidized at rates of 41%, 4.5%, and <0.2% compared to NOHA by iNOS, nNOS, and eNOS, respectively.4 The Km of 300 µM and kcat of 350 min−1 for iNOS gives a kcat/Km of 1.17, which is 9-fold lower than NOHA (kcat/Km = 10.6).4
