Sphingosine kinase (SPHK) is an important enzyme in the sphingolipid metabolic pathway. SPHKs phosphorylate sphingolipid D-erythro-sphingosine to yield sphingosine-1-phosphate (S1P). To date, two isoforms of SPHK, SPHK 1 and SPHK 2, have been identified and characterized from mammalian cells.1,2 SKs, through the formation of S1P, have been implicated in a diverse range of cellular processes, including cell proliferation, apoptosis, calcium homeostasis, angiogenesis, and vascular maturation.3 Thus SPHKs are potential therapeutic targets for the control of cancer, inflammation, and other diseases.4,3
