Description

Traditional purification of urine and plasma samples prior to measurement by EIA has required the use of reverse phase C-18 columns and various organic solvents. As this method relies upon polarity for separation, the final purified product is a non-specific mixture of many compounds. Further purification by TLC or HPLC is then required to isolate a single product. As an alternative, affinity chromatography provides a specific, reliable, and convenient means of purification for a variety of sample types. Cayman Chemical has developed an affinity sorbent for the purification of 8-isoprostane (8-iso PGF2α) from urine and plasma samples. The affinity sorbent can be regenerated by following a simple washing procedure, depending on the samples, please see product insert for details. Each vial of sorbent contains 1 ml of 8-isoprostane affinity sorbent (mouse anti-8-isoprostane covalently bound to Sepharose 4B). 8-Isoprostane is a potent pulmonary and renal vasoconstrictor1 and has been implicated as a causative mediator of hepatorenal syndrome and pulmonary oxygen toxicity.2 It has also been proposed as a marker of antioxidant activity. Cayman Chemical’s 8-Isoprostane EIA Kit (Catalog Nos. 10367 and 10368) is a highly sensitive and accurate assay, and our affinity purification reagents improve it even more.
