Low-density lipoproteins (LDL) is the major carrier of cholesterol in the blood, accounting for more than 60% of total plasma cholesterol. LDL is taken up by hepatic and extrahepatic tissues through LDL receptor-mediated endocytosis where it is internalized and degraded to free cholesterol and amino acids. Uptake of native and oxidized LDL by monocyte-derived macrophages in the vessel wall results in accumulation of cholesterol and formation of atherosclerotic plaques. The mechanism underlying the uptake of LDL by both hepatic tissue and cells of the cardiovascular system is not clearly understood. Thus, LDL uptake and its regulation are important therapeutic targets for atherosclerosis and related diseases. Cayman’s LDL Uptake Cell-Based Assay Kit provides a convenient tool for studying LDL uptake and regulation at cellular level. The kit employs human LDL conjugated to DyLight 549 as a fluorescent probe for detection of LDL uptake into cultured cells. A LDL receptor-specific polyclonal antibody and a DyLight 488-conjugated secondary antibody are included for identifying the distribution of LDL receptors.
