STEP: Surface Transfection and Expression Protocol

Stable cell lines are difficult to generate particularly when multiple expression constructs are involved. In addition, it is not possible to modulate the stoichiometry of transgene expression and the property of the stable cell line may change over time. Transient (solution-based) transfection has its advantages, but it is not compatible with many cell lines, and it has a limited co-transfection efficiency. STEP (Surface Transfection and Expression Protocol) technology has overcome many of the disadvantages of conventional transient and stable transfection approaches. It involves the formation of a transfection complex applied directly on the growing surface of culture vessels. This enhances both the transfection and co-transfection efficiencies in many cell lines and even primary cultures. The STEP complex is evenly coated on multi-well plates. The homogenous coating of STEP complex provides superior transfection efficiency consistently from well to well, which is essential in an assay for high throughput screening.
