Active regulator of SIRT1 (AROS) is a chaperone for ribosomal protein S19 (RPS19) and a modulator of sirtuin 1 (SIRT1) activity.1,2 It is an intrinsically disordered protein that contains localized α-helices.2 AROS is ubiquitously expressed and localizes to the nucleolus and cytoplasm.3,4 It is involved in ribosome biogenesis, specifically the assembly of the small subunit processome where it stabilizes RPS19, and protein synthesis.1,3 AROS activates or inhibits SIRT1 in a context-dependent manner.2,4,5 Methylation of AROS at lysine 27 by histone-lysine N-methyltransferase NSD2 increases the affinity of AROS for SIRT1, which subsequently activates SIRT1 to increase fatty acid β-oxidation in cancer cells.6 Increased expression of RPS19BP1, the gene encoding AROS, is associated with poor prognosis, higher tumor stage and size, microvascular and portal vein invasion, and increased α-fetoprotein levels in patients with non-cirrhotic hepatocellular carcinoma (HCC).7 Cayman’s AROS Polyclonal Antibody (aa 1-50) can be used for Western blot (WB).
