Giripladib is a cytosolic phospholipase A2 (cPLA2) inhibitor.1,2 It increases arachidonoyl-containing phosphatidylcholine levels in thrombin-stimulated isolated human platelets.1 Giripladib reduces platelet adhesion and accumulation in collagen-coated capillary tubes and inhibits thrombin- or convulxin-stimulated thromboxane B2 (TXB2) production in isolated human platelets. It reduces accumulation of PUFAs, malondialdehyde (MDA), and reactive oxygen species (ROS) in primary human vascular smooth muscle cells (VSMCs) stimulated with serum isolated from patients with chronic kidney disease (CKD) and inhibits ferroptosis in the same cells.2 Giripladib reduces atherosclerotic plaque vulnerability in an ApoE-/- mouse model of CKD.
