NLG-919 analog is an inhibitor of indoleamine 2,3-dioxygenase 1 (IDO1) and tryptophan 2,3-dioxygenase (TDO; IC50s = 79 and 25 nM, respectively, in cell-free assays).1 It decreases kynurenine levels in cells expressing human IDO1 (EC50 = 83 nM).2 NLG-919 analog (0.025-1 µM) decreases the percentage of anti-CD3- and anti-CD28-activated Jurkat T cells that are positive for programmed cell death protein 1 (PD-1).3 In vivo, NLG-919 analog (100 mg/kg) decreases tumor volume and weight and the percentage of intratumoral CD4+CD25+ regulatory T cells (Tregs) in a B16/F10 murine melanoma model when administered alone or in combination with paclitaxel (Item No. 10461).4
