Gallamine is an allosteric modulator of muscarinic acetylcholine receptors.1 It slows the dissociation of the muscarinic antagonist [3H]NMS from M1-M5 muscarinic receptors, with the highest potency for M2 receptors. Gallamine is cardioselective, inhibiting [3H]NMS binding in rat heart tissue homogenates more potently than in other tissues (IC50s = 0.8 and 3-30 µM, respectively).2 Formulations containing gallamine have previously been used as neuromuscular blockers during surgery.
