NPD4928 is an inhibitor of ferroptosis suppressor protein 1 (FSP1) and an enhancer of ferroptosis.1 It binds to FSP1 and inhibits its activity in a cell-free NADH consumption assay. NPD4928 (3 µM) enhances cytotoxicity induced by the ferroptosis inducer RSL3 (Item No. 19288) in PANC-1 prostate cancer cells in a synergistic manner. It also enhances cytotoxicity induced by sulfasalazine (Item No. 15025) and L-buthionine sulfoximine (BSO) in the same cells. NPD4928 in combination with RSL3 induces cytotoxicity in PC3, MKN74, DU145, MIA PaCa-2, and A549 cancer cells (IC50s = 105, 95, 103, 140, and 155 nM, respectively) but not BxPC-3 or RPMI-8226 cancer cells (IC50s = >30 µM for both).
