HUP-55 is an inhibitor of prolyl endopeptidase (IC50 = 5 nM).1 It reduces α-synuclein dimerization in Neuro2a cells, induces autophagy in HEK293 cells, and decreases hydrogen peroxide-induced increases in reactive oxygen species (ROS) production in SH-SY5Y cells when used at a concentration of 10 µM. HUP-55 (10 mg/kg) decreases ipsilateral paw use and striatal oligomeric α-synuclein levels in a mouse model of Parkinson’s disease induced by adenoviral delivery of an α-synuclein-encoding vector (AAV-αSyn).
