DOWNLOAD NOWD359-0396 is an inhibitor of the NOD-like receptor protein 3 (NLRP3) inflammasome.1 It is selective for NLRP3 over the absent in melanoma 2 (AIM2) and NLR family CARD domain-containing protein 4 (NLRC4) inflammasomes at 100 µM. D359-0396 inhibits pyroptosis induced by ATP (Item Nos. 14498 | 40182) or nigericin (Item No. 11437) in LPS-primed primary mouse bone marrow-derived macrophages (BMDMs). It also inhibits LPS- and ATP-induced NLRP3 self-oligomerization and apoptosis-associated speck-like protein (ASC) oligomerization in vitro at 20 µM. D359-0396 decreases disease severity in a mouse model of experimental autoimmune encephalomyelitis (EAE) and increases survival in a mouse model of septic shock.
