SMU-L11 is a toll-like receptor 7 (TLR7) agonist.1 It selectively activates TLR7 over TLR8 signaling in a cell-based reporter assay (EC50s = 0.024 and 4.9 µM, respectively) and is inactive at TLR2, -3, and -4 up to 100 µM. SMU-L11 enhances the protein-protein interaction between TLR7 and MyD88 and increases the phosphorylation of p65, p38, JNK, ERK1/2, and IFN regulatory factor 7 (IRF7) in isolated mouse bone marrow-derived dendritic cells (BMDCs). SMU-L11 increases TNF-α and IL-6 levels in RAW 264.7 macrophages and isolated mouse BMDCs and TNF-α and IL-1β levels in isolated human peripheral blood mononuclear cells (PBMCs). SMU-L11 increases the number of splenic and tumoral natural killer (NK) cells, CD4+ and CD8+ T cells, and M1-like macrophages and reduces tumor growth in a B16/F10 murine melanoma model.
