(E)-3-(2-Fluorophenyl)-N-(1H-indol-5-yl)acrylamide is an inhibitor of monoamine oxidase B (MAO-B) and an inducer of nuclear factor erythroid 2-related factor 2 (Nrf2).1 It inhibits MAO-B (IC50 = 3.61 µM) and increases levels of Nrf2 in a reporter assay using AREc32 cells (IC50 = 0.361 µM). (E)-3-(2-Fluorophenyl)-N-(1H-indol-5-yl)acrylamide scavenges peroxyl radicals in an oxygen radical absorbance capacity (ORAC) assay (IC50 = 2.77 µM) and inhibits LPS-induced increases in nitrite levels in BV-2 microglial cells (EC50 = 11.6 µM). It inhibits 6-OHDA- or rotenone- and oligomycin A-induced decreases in viability in SH-SY5Y neuroblastoma cells when used at a concentration of 1 µM.
