Mc-Val-Ala-PAB is a peptide linker molecule that has been used in the synthesis of antibody-drug conjugates (ADCs).1,2 It contains a maleimidocaproyl (Mc) group that can be conjugated to an antibody and a p-aminobenzyl (PAB) spacer that allows the peptide to be linked to active compounds, such as anticancer agents or antibiotics. Mc-Val-Ala-PAB cleavage in vitro and in vivo by cathepsin B allows the active compound to be released at the target site. It has been used in the synthesis of ADCs containing the antimitotic compound monomethyl auristatin E (MMAE; Item No. 16267) or the antibiotic KRM-1657.
