ABD957 is an inhibitor of α/β-hydrolase domain-containing protein 17 (ABHD17; IC50 = 0.21 µM for ABHD17B).1 It is selective for ABHD17A-C over a panel of 52 serine hydrolases, including the depalmitoylases ABHD10, palmitoyl-protein thioesterase 1 (PPT1), and lysophospholipase 1 (LYPLA1) and LYPLA2, at 10 µM but does inhibit ABHD6, ABHD11, ABHD13, and carboxylesterase 1 (CES1) and CES2 at the same concentration. ABD957 increases the palmitoylation of N-Ras in OCI-AML-3 acute myeloid leukemia (AML) cells expressing GFP-N-RasG12D (EC50 = 29 nM). It also increases the plasma membrane localization of N-Ras in the same cells when used at a concentration of 500 nM. ABD957 inhibits the growth of OCI-AML-3 cells in a concentration-dependent manner.
