DOWNLOAD NOWHyNic NHS ester is a heterobifunctional linker molecule.1 It contains an amine-reactive N-hydroxysuccinimide (NHS) moiety and a hydrazine moiety that reacts with the aldehyde functional group in SFB (succinimidyl 4-formylbenzoate; Item No. 30477) to form a bis-aryl hydrazone linker that is quantifiable spectrophotometrically at 354 nm. HyNic NHS ester has been used in the synthesis of an antibody-drug conjugate (ADC) that contains the toll-like receptor 7 (TLR7) agonist UC-1V150 and the anti-CD20 monoclonal antibody rituximab and increases pro-inflammatory activity in RAW 264.7 macrophages.2 HyNic NHS ester has also been used in the formation of polymer-enzyme, protein-liposome, and peptide-peptide conjugates.1,3,4
