Ro 5-3335 is a benzodiazepine that modulates the protein-protein interaction between RUNX family transcription factor 1 (RUNX1) and core-binding factor subunit β (CBFβ).1 It inhibits transactivation of the CSF-1 receptor (CSF-1R) promoter, a target of RUNX1 and CBFβ, in a reporter assay. Ro 5-3335 reduces viability of leukemia cells harboring CBF or RUNX1 fusion proteins (IC50s = 1.1-21.7 µM) to a greater extent than non-CBF leukemia cells (IC50s = 54.6->200 µM). It also inhibits RUNX1/CBFβ-dependent hematopoiesis in zebrafish embryos.
