RS 67333 is an agonist of the serotonin (5-HT) receptor subtype 5-HT4.1 It selectively binds to 5-HT4 (Ki = 2 nM) over 5-HT1A, 5-HT2A, 5-HT2C, and 5-HT3 receptors (Kis = 398, 501, 794, and 398 nM, respectively), as well as α- and β-adrenergic and muscarinic receptors (Kis = >200 nM for all). It also binds to σ1- and σ2-receptors (Kis = 1 and 10 nM, respectively). RS 67333 induces relaxation of carbachol-precontracted rat esophageal muscularis mucosae preparations (EC50 = 2 nM) and induces tachycardia in anesthetized micropigs at 4.9 µg/kg. It reverses scopolamine-induced memory deficits in mice in the Y-maze test when administered at a dose of 1 mg/kg.2
