PDS-0330 is an inhibitor of the protein-protein interaction between claudin-1 and c-Src.1 It binds to claudin-1 (Kd = 22.9 µM) and reduces the levels of phosphorylated c-Src and Akt in SW620 colorectal cancer cells. PDS-0330 decreases the viability of SW620 cells, which endogenously overexpress claudin-1, but not of HCT116 colorectal cancer cells, which do not express claudin-1 (EC50s = 9.4 and 95 µM, respectively). It induces apoptosis in, and inhibits the migration of, SW620 cells. PDS-0330 (5-10 mg/kg) increases intratumoral apoptosis and reduces tumor volume in a SW620 mouse xenograft model and in patient-derived xenograft (PDX) mouse model of chemotherapy-resistant colon cancer.
