Preloaded and Loadable LNPs eliminate the need for expensive and complex mixing steps, ensuring ease of use. The LipidLaunch™ LNP Formulation Buffer is an acidic aqueous buffer designed to optimally encapsulate nucleic acid cargo within LNPs. It consists of an RNase-free solution of 50 mM sodium acetate at pH 4.5. The LipidLaunch™ LNP Formulation Buffer can be used with any selected lipid mixture, including LipidLaunch™ Exploration and Uptake Kits as well as LipidLaunch™ Ionizable Lipid LNP Screening and Helper Lipid Sets.
The acid dissociation constant (pKa) is the pH at which the ionized and nonionized forms of an ionizable lipid exist in equal concentrations. pKa impacts the LNP encapsulation efficiency, efficacy, delivery, and toxicity and is an important parameter in LNP characterization.
