OBESITY RESEARCH SOLUTIONSO-304 is a pan-activator of AMP-activated protein kinase (AMPK) with mitochondrial uncoupling activity.1,2 It increases the levels of phosphorylated AMPK (pAMPK) in various primary human cells expressing AMPKβ1 or AMPKβ2 and increases mitochondrial respiration in C2C12 myotubes in a concentration-dependent manner. O-304 (100 mg/kg) prevents the development of insulin resistance in diet-induced obese (DIO) mice.1 It stimulates insulin-independent glucose uptake and utilization in the skeletal and cardiac muscle of diabetic mice.2 It also reverses hyperinsulinemia and insulin resistance, improves cardiac function, and increases exercise capacity in aged mice at 0.5 mg/g.3
