BMS 509744 is an inhibitor of IL-2-inducible T cell kinase (Itk; IC50 = 19 nM).1 It is selective for Itk over 18 other kinases (IC50s = >1,100 nM for all). BMS 509744 inhibits IL-2 secretion from Jurkat T cells and isolated human peripheral blood mononuclear cells (PBMCs) induced by antibodies targeting CD3 (IC50s = 0.25 and 0.39 µM, respectively). It decreases serum levels of Il-2 and the lung infiltration of total immune cells and eosinophils in a mouse model of ovalbumin-induced lung inflammation when administered at a dose of 25 mg/kg. Topical application of BMS 509744 (3%) reduces ear skin thickness and decreases the number of infiltrating immune cells in a mouse model of imiquimod-induced skin inflammation.2
