Fz7-21 is a peptide antagonist of the frizzled family G protein-coupled receptor 7 (FZD7; IC50 = 100 nM).1 It inhibits Wnt3a-induced β-catenin signaling in a reporter assay using HEK293 cells. Fz7-21 (100 µM) reduces the number of cells positive for leucine-rich α-2-glycoprotein 5 (Lrg5), a marker of intestinal stem cells, and the mRNA encoding achaete-scute homolog 2 (Ash-2) and Axin2 and increases mRNA encoding mucin 2 (Muc2) in primary mouse intestinal organoids.
