T100-Mut is a cell-permeable peptide and transient receptor potential ankyrin 1 (TRPA1) inhibitor that contains the C-terminal sequence of Tmem100-3Q, a mutant form of the endogenous peptide Tmem100.1 It reduces calcium flux induced by mustard oil, a TRPA1 agonist, in primary mouse dorsal root ganglia (DRG) when used at a concentration of 200 nM. Subcutaneous administration of T100-Mut (5 µl of a 2 mM solution) into the hind paw reduces paw licking induced by mustard oil but not the TRP vanilloid 1 (TRPV1) agonist capsaicin (Item Nos. 92350 | 10010743) in mice. It also reduces mechanical hyperalgesia induced by complete Freund’s adjuvant (CFA) or paclitaxel (Item No. 10461) in wild-type, but not TRPV1 knockout, mice.
