S7 is a peptide antagonist of the IL-6 receptor.1,2 It inhibits IL-6 binding to the IL-6 receptor in a concentration-dependent manner.1 S7 (50 µM) inhibits IL-6-induced increases in VEGF levels in C-33 A cervical cancer cells and RPMI-8226 B cell lymphocytes. It reduces tumor volume in an IL-6-overexpressing C-33 A cervical cancer mouse xenograft model when administered at a dose of 50 mg/kg every two days. When coupled to cysteine and the surface of lipid nanoparticles (LNPs) encapsulating doxorubicin, S7 increases LNP glioma targeting and increases survival in a U251 glioblastoma mouse xenograft model. 2
