TCN 237 is an antagonist of NR2B subunit-containing NMDA receptors (Ki = 0.85 nM).1 It selectively inhibits glutamate and glycine-induced currents in whole-cell patch-clamp assays using L-M(TK-) cells expressing NR2B subunit-containing NMDA receptors over L-M(TK-) cells expressing NR2A- or NR2D subunit-containing NMDA receptors and HEK293 cells expressing NR2C subunit-containing NMDA receptors. TCN 237 (3 mg/kg) inhibits carrageenan-induced mechanical hyperalgesia in rats.
