VcMMAE is a drug-linker molecule that has been used in the synthesis of antibody-drug conjugates (ADCs).1,2 It contains maleimide, the cathepsin B substrate valine-citrulline (Val-Cit) dipeptide, and p-aminobenzyl carbamate (PABC) conjugated to the antimitotic agent monomethyl auristatin E (MMAE).1 The maleimide in VcMMAE allows it to be conjugated to cysteine residues in antibodies, which target the ADC to specific cell types. Once inside the cell, the Val-Cit dipeptide is cleaved by cathepsin B and MMAE is released.3,1 Formulations containing VcMMAE have been used in the treatment of diffuse large B cell lymphoma (DLBCL), Hodgkin lymphoma, systemic anaplastic large cell lymphoma, and urothelial cancer. Cayman’s VcMMAE Polyclonal Antibody can be used in ELISA for the detection of ADCs containing the VcMMAE linker.
