    Perfringolysin O (PFOC459A) is a recombinant mutant form of PFO that permeabilizes the cellular plasma membrane to form 25-30 nm pores, allowing the diffusion of membrane-impermeable substrates, inhibitors, and other biomolecules into live cells.1-3 As a cholesterol-dependent cytolysin, PFOC459A preferentially permeabilizes the plasma membrane while preserving mitochondrial integrity and function, enabling functional mitochondrial studies in an intact cellular context without organelle isolation. Cayman’s Perfringolysin O (C459A mutant; C. perfringens, recombinant) protein can be used in permeabilized cell assays to evaluate mitochondrial oxygen consumption, drug mechanism of action, metabolic shifts to various stimuli, comparison of healthy and disease states, or assessment of bioprocessing and manufacturing-induced cellular stress.

    For use in cell-based assays, including Cayman’s Extracellular Oxygen Consumption Assay Kit (Item No. 502880), it is important to identify an optimal concentration of PFOC459A that sufficiently permeabilizes the plasma membrane. An example protocol can be found below but must be adapted for each experimental system and readout.
