BRD4780 is an antagonist of the imidazoline I1 receptor (Ki = 42 nM).1 It is selective for the imidazoline I1 receptor over α1-, α2A-, α2B-, and α2C-adrenergic receptors (Kis = >100, >20, >20, and >20 µM, respectively). BRD4780 also binds to transmembrane P24 trafficking protein 9 (TMED9) in a thermal shift assay and releases mucin 1-fs (MUC1-fs), a mutant form of MUC1 caused by a frameshift mutation in MUC1, from TMED9-containing vesicles in kidney cells isolated from patients with MUC1 kidney disease.2 In vivo, BRD4780 (1, 10, and 50 mg/kg) reduces renal MUC1-fs accumulation in transgenic mice heterozygous for human MUC1-fs.
