UNC9426 is an inhibitor of the TAM family kinase Tyro3 (IC50 = 2.1 nM).1 It is selective for Tyro3 over MerTK and Axl (IC50s = 580 and 190 nM, respectively) and 74 other kinases in a panel at 1 µM. UNC9426 (10 µM) reduces ADP- or collagen-induced platelet aggregation in isolated human platelet-rich plasma. It does not increase total bleeding time in mice after tail tip transection when administered at a dose of 10 mg/kg. UNC9426 also reduces the uptake of phosphatidylserine-coated microparticles, a model of apoptotic cells, by THP-1 macrophages in an efferocytosis assay in a concentration-dependent manner and reduces colony formation in NCI H1650 or H1975 non-small cell lung cancer (NSCLC) cells (EC50s = 170 and 197 nM, respectively).
