CMPD-39 is an inhibitor of ubiquitin-specific protease 30 (USP30; IC50 = 20 nM in a cell-free assay).1 It is selective for USP30 over a panel of 39 other deubiquitinases at 10 µM.2 CMPD-39 increases the number and size of mitolysosomes in SH-SY5Y cells and increases pexophagy in U2OS cells. It restores mitophagy in dopaminergic neurons derived from Parkinson's disease patient-derived induced neuronal progenitor cells that were generated from fibroblasts harboring a loss of function mutation in PRKN.
