GLPG3970 is an inhibitor of salt-inducible kinase 1 (SIK1), SIK2, SIK3, and receptor-interacting serine/threonine kinase 2 (RIPK2; IC50s = 282.8, 7.8, 3.8, and 78.4 nM, respectively).1 It is selective for SIK1, SIK2, SIK3, and RIPK2 over a panel of 371 other kinases at 1 µM. GLPG3970 reduces the levels of TNF-α and IL-12 and increases the level of IL-10 in LPS-stimulated primary human monocytes and whole blood. It decreases disease severity in a mouse model of colitis induced by dextran sodium sulfate (DSS) when administered at a dose of 30 mg/kg.
