AZD 9574 is a poly(ADP-ribose) polymerase 1 (PARP1) inhibitor (IC50 = 0.005 µM).1 It is selective for PARP1 over PARP2 (IC50 = >93 µM) and induces synthetic lethality in DLD-1 colorectal cancer cells lacking BRCA2 (IC50 = 0.016 µM). AZD 9574 (10-30 mg/kg) reduces tumor volume but not body weight in a BRCA2-/- DLD-1 mouse xenograft model. It has been used as an adjuvant host-directed therapy (HDT) that, when administered with isoniazid (Item No. 20378), increases survival and reduces neurological deficits in a mouse model of M. tuberculosis meningitis.2
