XL177A is an irreversible inhibitor of ubiquitin-specific protease 7 (USP7; IC50 = 0.34 nM).1 It is selective for USP7 over a panel of 41 deubiquitinating enzymes at 1 µM. XL177A decreases murine double minute 2 (MDM2) levels, increases p53 and p21 levels, and induces cell cycle arrest at the G1 phase in MCF-7 breast cancer cells. It decreases tumor volume in an NB-SD neuroblastoma mouse xenograft model at 75 mg/kg.2 XL177A also decreases mRNA expression of the genes encoding angiotensin-converting enzyme 2 (ACE2) and dipeptidyl peptidase 4 (DPP-4) in Huh7.5 cells and viral titers in BV-2 cells infected with the coronavirus mouse hepatitis virus strain 3 (MHV-3).3
