Tacaciclib is a covalent inhibitor of cyclin-dependent kinase 7 (Cdk7; IC50s = 12, 5, and 0.9 nM after 20, 60, and 180 minutes, respectively).1 It reduces the viability of myeloid blasts isolated from patients with primary or relapsed/refractory acute myeloid leukemia (AML; mean IC50s = 0.33 and 0.3 µM, respectively). Tacaciclib reduces the levels of c-Myc and increases the levels of p53 and p21 in AML cell lines and acts synergistically with venetoclax (ABT-199; Item No. 16233) to reduce the viability of venetoclax-resistant MOLM-13 cells. It induces cell cycle arrest at the G1 or G2/M phase in MOLM-13 cells at 0.5 or 2 µM, respectively. Tacaciclib (60 mg/kg) reduces the percentage of human CD45+ cells in the bone marrow, indicating reduced leukemia cell burden, and increases survival in an orthotopic MOLM-13 mouse xenograft model.
