OBESITY RESEARCH SOLUTIONSAZD 3988 is an inhibitor of diacylglycerol acyltransferase 1 (DGAT-1; IC50 = 0.6 nM for the human enzyme).1 It is selective for DGAT-1 over DGAT-2 at 10 µM. AZD 3988 inhibits triacylglycerol synthesis in HuTu 80 cells (IC50 = 0.5 nM) and oleic acid-induced lipid droplet accumulation in human duodenal organoids at 100 nM.1,2 It decreases plasma triglyceride levels in an oral lipid tolerance test in fasted rats. AZD 3988 (20 mg/kg, p.o.) reduces body weight in diet-induced obese (DIO) mice.1
