OBESITY RESEARCH SOLUTIONSTM-25659 is a modulator of transcriptional coactivator with PDZ-binding motif (TAZ).1 It increases nuclear localization of TAZ in C3H/10T1/2 cells and reduces differentiation of 3T3-L1 mouse preadipocytes mediated by peroxisome proliferator-activated receptor γ (PPARγ) at 100 µM. It also increases C3H/10T1/2 osteoblast differentiation at 50 µM. TM-25659 (50 mg/kg) reduces weight gain in a mouse model of high-fat diet-induced obesity. In addition, it reduces weight gain and partially restores bone mineral density in a rat model of ovariectomy-induced bone loss.
