Necrostatin-34 is an inhibitor of receptor-interacting serine/threonine kinase 1 (RIPK1; IC50 = 5.5 µM).1 It inhibits TNF-α-induced necroptosis in Jurkat cells lacking Fas-associated death domain protein (FADD) and L-929 cells (IC50s = 6.67 and 1.35 µM, respectively). Necrostatin-34 (10 µM) inhibits necroptosis induced by the chemical inducer of dimerization for FK506 binding protein (FKPB) fusion proteins AP20187 (Item No. 36808) and the pan-caspase inhibitor Z-VA-DL-D(OMe)-FMK (zVAD-FMK; Item No. 27421) in L-929 cells expressing an FKBP and RIPK1 fusion protein.
