    Product Description

    This assay utilizes proprietary mammalian reporter cells expressing the Aryl Hydrocarbon Receptor (AhR; AHR). The principal application of this AhR bioassay is to screen processed environmental samples, with the aim of quantifying the aggregate bioactivities, if any, of contaminating biohazardous chemical entities. Contaminants of particular concern include the polycyclic aromatic hydrocarbons (PAHs) and halogenated aromatic hydrocarbons (HAHs) such as dioxin, dioxin-like chemicals, and PCBs.

    INDIGO's Reporter Cells contain a luciferase reporter gene functionally linked to an AhR-responsive promoter. Thus, quantifying changes in luciferase expression in the treated reporter cells provides a sensitive surrogate measure of the changes in AhR activity.

    INDIGO’s Reporter Cells are transiently transfected and prepared as frozen stocks using a proprietary CryoMite™ process. This cryo-preservation method allows for the immediate dispensing of healthy, division-competent reporter cells into assay plates. There is no need for intermediate treatment steps such as spin-and-rinse of cells, viability determinations or cell titer adjustments prior to assay setup.

    This assay kit provides the convenience of an all-inclusive cell-based reporter assay system for AhR activity. In addition to Reporter Cells, included are two optimized media for use during cell culture and in diluting the user's test samples, Luciferase Detection Reagent, and a cell culture-ready assay plate.
