HRX215 is an inhibitor of MAP kinase kinase 4 (MKK4; IC50 = 0.02 µM).1 It is selective for MKK4 over JNK1, B-RAF, and MKK7 (IC50s = 7.07, 11.46, and 14.97 µM, respectively). HRX215 (10 mg/kg) decreases hepatocyte apoptosis in a mouse model of liver damage induced by carbon tetrachloride (CCl4). It increases survival and hepatic size and regeneration, decreases intracranial pressure (ICP), and reduces blood ammonia, total bilirubin, and aspartate aminotransferase (AST) levels in a porcine model of partial hepatectomy-induced liver failure when administered at 5 mg/kg.
