Explore CGMP Lipid Services(R)-C12-200 is an ionizable cationic lipid (pKa = 7.25) and isomer of C12-200 (Item No. 36699) that has been used in the formation of lipid nanoparticles (LNPs) for the delivery of mRNA in vivo.1 Intravenous administration of LNPs containing (R)-C12-200 and encapsulating mRNA encoding Cre recombinase increases the percentage of tdTomato+ hepatocytes, endothelial cells, and Kupffer cells in the liver of Ai14 mice to a lesser extent than LNPs containing (S)-C12-200 (Item No. 38885) and encapsulating Cre recombinase. LNPs containing (R)-C12-200 and encapsulating mRNA encoding purine nucleoside phosphorylase (PNP), an E. coli enzyme that converts fludarabine phosphate (Item No. 14251) into the cytotoxic agent 2-fluoroadenine, induce intratumoral expression of PNP and reduce tumor volume in a FaDu head and neck squamous cell carcinoma (HNSCC) mouse xenograft model when co-administered with fludarabine phosphate.2
