P-2281 is an mTOR inhibitor.1 It inhibits serum-induced phosphorylation of p70 ribosomal S6 kinase 1 (p70S6K1) in serum-starved HCT116 colon cancer cells at 30 µM and serum-induced phosphorylation of 4E-BP1 in serum-starved H460 non-small cell lung cancer (NSCLC) cells at 3 or 10 µM. P-2281 (30 µM) reduces concanavalin A-induced IFN-γ production but not LPS-induced TNF-α production in isolated human peripheral blood mononuclear cells (PBMCs). It decreases disease severity in a mouse model of colitis induced by dextran sulfate sodium (DSS) at 15 mg/kg.
