Neutralizing Antibody Analysis Post COVID-19 Infection or Vaccination

Cayman's Bioanalysis & Assay Development Services partnered with GenScript to examine the onset and durability of the COVID-19 vaccine-induced neutralizing antibody response.

The ability to detect antibodies that neutralize SARS-CoV-2 and monitor seroprevalence is paramount to determining the efficacy and longevity of COVID-19 vaccines. Cayman scientists helped to validate a simple 96-well plate-based assay that provides accurate SARS-CoV-2 population surveillance analysis and reveals the presence of antibodies in recovered or vaccinated individuals that block the interaction between the SARS-CoV-2 receptor binding domain and the ACE2 receptor. A full analysis of neutralizing antibody responses from individuals who received either the Pfizer, Moderna, or Johnson & Johnson vaccines using this assay has been published in the journal Vaccine.

Using GenScript's cPass™ sVNT Assay Kit, Cayman scientists partnered with GenScript to explore the onset and durability of the neutralizing antibody response following vaccination. Semi-quantitative, longitudinal detection of neutralizing antibodies in four subjects receiving the Pfizer vaccine show low or undetectable levels until the second dose where levels rose rapidly and peaked within days following the second dose (see figure below). After this peak, a sharp initial decline in neutralizing antibodies was noted, and this decline tailed off approximately 30 days after the second dose.

Our scientists have also developed a research use only SARS-CoV-2 Neutralizing Antibody Detection ELISA Kit to detect any circulating neutralizing antibodies—independent of isotype—that block the interaction between the SARS-CoV-2 spike glycoprotein RBD and the ACE2 receptor. Our competitive ELISA includes a SARS-CoV-2 neutralizing antibody as a standard that was carefully tested to ensure its interference with the ACE2 protein for binding sites on the SARS-CoV-2 spike S1 RBD.
